ABSTRACT

Objective:To evaluate the efficacy and safety of Hymagic-4D multi-essence (miniHA, acetylated sodium hyaluronate, sodium hyaluronate, sodium hyaluronate cross-linked polymer) on sensitive skin.Methods:The clinical trial was conducted from April 2, 2019 to April 30, 2019 in the Cosmetic Dermatology Laboratory of the Third Affiliated Hospital of Sun Yat-Sen University. A total of 33 subjects (average age of 43±9.5) completed the trial. Subjects diagnosed as sensitive skin according to the criteria were enrolled. Hymagic-4D were randomly applied on one side of the face while macromolecular hyaluronic acid was applied on opposite side for 28 days. Skin biophysical properties, lactic test, clinical efficacy and safety were evaluated by the investigators at day 0, day 7, day 14 and day 28. Skin water content, TEWL and a*value were measured at the same time.Results:Dates showed that at day 7, day 14 and day 28, the a* values on Hymagy-4D using side were 8.54±3.08, 8.87 ±3.21 and 8.39±3.21, lower than that on the side of control 9.48±3.09, 9.51±3.30 and 9.03±2.95 ( P<0.01); skin roughness score, dryness score and erythema score on hymagy-4d using side were significantly lower than before ( P<0.01), and significantly lower than that on the control side ( P<0.01). Hydration, TEWL, pH value, L value and total score of lactic acid tingling test which were significantly improved on both facial skin compared to the baseline ( P<0.05) showed no statistical difference between two facial sides ( P>0.05) during 28-day treatment. Conclusions:This study demonstrates that Hymagic-4D has more effective in repairing skin barrier and inhibiting skin inflammation than single component hyaluronic acid on sensitive skin persons.

ABSTRACT

Objective:To investigate the level of regulatory T (Treg) cells of peripheral blood and evaluate the correlation between Treg cells and disease activity in rheumatoid arthritis (RA) to further clarify the role of Treg cells in the pathogenesis of RA.Methods:The participants included 300 patients with RA, from the Second Hospital of Shanxi Medical University between January 2016 and September 2018 and 100 gender and age matched healthy individuals. The absolute numbers and proportion of Treg cells and helper T (Th) 17 cells and the ratio of Th17/Treg in peripheral blood of these individuals were detected by flow cytometry combined with standard absolute counting beads. Main statistical analysis methods were t test, spearman correlation and rank sum test. Results:Patients with RA had lower absolute numbers [(35±13) cells/μl vs (26±17) cells/μl, t=4.815, P<0.01] and percentage [(5.1±1.5)% vs (4.4±2.0)%, t=4.111, P<0.01] of Treg cells compared with those in healthy controls, while the number ( t=1.114, P=0.266) and proportion ( t=1.577, P=0.116) of Th17 cells were not significantly changed. RA patients, regardless of new onset (no prior treatment) or treatment with immunosuppressants, had significantly lower absolute numbers of Treg cells [healthy controls (35±13) cells/μl; the Treg cells in new onset patients was (31±17) cells/μl, t=1.974, P=0.049; Treg cells in treated patients was (24±16) cells/μl, t=5.978, P<0.01] but not Th17 cells than healthy controls. With the increase of disease activity score uses 28 joint counts (DAS28), the absolute counts of peripheral Treg cells trended to decrease [remission group (33±15) cells/μl, low activity group (31±17) cells/μl, moderate activity group (28±17) cells/μl, high activity group (24±16) cells/μl], and were negatively correlated with DAS28( r=-0.139, P=0.016) and erythrocyte sedimentation rate (ESR) ( r=-0.116, P=0.046). Conclusion:The absolute counts of peripheral Treg cells decreased significantly, which are associated directly with the pathogenesis of RA rather than high-dose glucocorticoids or immunosuppressive agents. Changes in circulating Treg cells number may play a prominent role in the disease process of RA, which provides clinical basis for evaluation of disease activity and exploration of new effective therapies.

ABSTRACT

Objective@#To evaluate the effects of repeated ultraviolet A (UVA) radiation on DNA damage, repair and replication processes in human skin fibroblasts, and to explore their mechanisms.@*Methods@#Fibroblasts were isolated from the circumcised foreskins of 3 children in the Department of Urological Surgery, Third Affiliated Hospital, Sun Yat-sen University, and subjected to a primary culture. Cultured human skin fibroblasts of 3rd-10th passages were divided into 2 groups: UVA group treated with repeated UVA radiation to establish a chronic photodamaged cell model, and control group receiving no treatment. Cell counting kit 8 (CCK-8) assay, β-galactosidase staining and flow cytometry were performed to assess cellular proliferative activity, and determine the proportion of photoaged cells and apoptosis rate respectively. Reverse transcription reaction was performed to establish a differentially expressed cDNA library, which was then subjected to high-throughput sequencing. The cDNA sequencing results were compared between the control group and UVA group, and the differentially expressed genes were analyzed in Kyoto Encyclopedia of Genes and Genomes (KEGG) database. The data were compared between the two groups by using two independent sample t test.@*Results@#Compared with the control group, the UVA group showed significantly decreased cellular proliferative activity (72.0% ± 5.2% vs. 96.0% ± 3.7%, t = 6.51, P < 0.05) , but significantly increased proportion of photoaged cells (79.7% ± 5.2% vs. 6.4% ± 0.8%, t=24.12, P < 0.05) and apoptosis rate (29.0% ± 3.3% vs. 6.0% ± 5.9%, t= 5.89, P < 0.05) . Among the key enzymes involved in DNA mismatch repair, replication and base excision repair processes, the expression of DNA ligase 1 (Lig1) , ribonuclease (RNase) H2A and helicase Dna2 in the UVA group was 0.47 ± 0.13, 0.44 ± 0.07 and 0.49 ± 0.11 times (all P < 0.01) that in the control group respectively. After the UVA-induced chronic photodamage in the human skin fibroblasts, After the UVA-induced chronic photodamage in the human skin fibroblasts, Lig expression decrease could block DNA single-base excision repair, Lig and Lig1 expression decrease could block DNA multiple-base excision repair, Lig1 expression decrease could block DNA mismatch repair, and RNaseH2A, Dna2 and Lig1 expression decrease could block DNA replication.@*Conclusion@#Repeated UVA radiation can change the expression of key enzymes involved in DNA base excision repair, DNA mismatch repair and DNA replication processes in skin fibroblasts, and then affect DNA repair and DNA replication processes in skin fibroblasts.

ABSTRACT

Objective To investigate the effects of remifentanil in painless bronchoscopic examination of elderly panents.Methods One hundred and twenty elderly patients undergoing painless bronchoscopic examination were randomly divided into 4 groups (n=30),the group A was administered with propofol and the groups B,C and D were administered with propofol and remifentanil(remifentanil dose:0.05 μg · kg-1 · min-1 in the group B,0.10μg · kg-1 · min-1 in the group C and 0.15 μg · kg-1 · min-1 in the group D).The body moving response,hemodynamic changes,propofol dose,examination time,wake-up time,departure time and adverse reactions in each group were observed during the examination process.Results During the examination process,the body moving response and bucking of the group A were significantly higher than those of the other three groups(P<0.05);in hemodynamics,the group A fluctuated greatly,while the group B,C and D were relatively stable,but the incidence rate of hypotension,bradycardia and transient respiratory depressionof the group D were higher than those of the other three groups(P<0.05);propofol dose of the group A was significantly higher than that of the other three groups (P<0.05)and the group B was higher than the group C and D(P<0.05).In wake-up time and departure time,the groups B,C,D were significantly better than the group A(P<0.05),while the group C and D were better than the group B(P<0.05).Conclusion Remifentanil can be used in painless bronchoscopic examination for elderly patients,and the effect is better by using propofol combined with remifentanil at a dose of 0.1 μg · kg-1 · min-1.

The metabolism of thyroid hormone changes during pregnancy. Subclinical hypothyroidism during pregnancy may adversely affect the pregnancy outcomes and neuropsychological development of child, controversially. The diagnostic criteria of subclinical hypothyroidism during pregnancy need to be specific. It is important to figure out when should pregnancy with subclinical hypothyroidism be treated and what is the specific goal of the treatment.

ABSTRACT

Objective To apply Six-minute Walk Test (6MWT) in acute stroke patients and analyze the results. Methods From Novem-ber, 2014 to January, 2015, 29 acute stroke patients with multiple or single infarction accepted 6MWT. The distance, distance per minute, blood pressure and heart rate before and after test, adverse events or discomforts during the test were recorded. Weak and fatigue scores were calculated. Results The mean of distance in acute stroke patients was (240.1±17.2) m, that was (46.8±3.3)%of the predicted values in the healthy. Male patients walked (255.4±21.40) m, not different from (214.9±28.4) m in female (t=-1.151, P=0.26). The distance liked to be negatively correlated with age (r=-0.356, P=0.058). The distance was similar in each minute. Weak score was (46.8±3.3)%, fatigue score was mostly between-20 and 20, and no correlation was found between them. Blood pressure and heart rate increased significantly at the end of the test (t>2.476, P<0.05). Few of patients complained mild discomforts. Conclusion 6MWT is safe and suitable for acute stroke patients for physical mobility judgement, but adverse events need to be cared.

ABSTRACT

Objective To analyze the radiographic characteristics of right hand X-ray film of adult patients with Kaschin-Beck disease (KBD) in Qinghai Province, to understand the most affected locations in adult KBD. Methods According to the criteria of KBD diagnose (WS/T 207-2010), 111 cases of patients were taken X-ray films of right hands. Joint space narrow, joint deformity, subchondral sclerosis, osteophyte, coarse and irregularity of joint, marginal retraction sign and capsule changes were chosen as the descriptive indexes, and these indexes were analyzed with SPSS 17.0. Results A total of 111 cases adult patients with KBD were examined right hand by X-ray, abnormality on X-ray film were 103 cases, the abnormal rate was 92.79%. The most affected fingers were Ⅱ- Ⅳphalanx bones, Ⅱphalanx bones accounted for about 92.23% (95/103), Ⅲ phalanx bones accounted for about 99.03% (102/103), and Ⅳ phalanx bones accounted for about 99.03% (102/103). There was significant difference of the abnormality between th e proximal phalanx and the middle phalanx among the Ⅱ - Ⅳ phalanx bones(χ2=79.33, P<0.05). Abnormal numbers of joint deformity, marginal retraction sign, coarse and irregularity of joint, osteophyte, capsule changes and joint space narrow in the proximal phalanx were 212, 7, 134, 47, 15 and 115 in Ⅱ - Ⅳ proximal phalanx, respectively; while the abnormal numbers of joint deformity, marginal retraction sign, coarse and irregularity of joint, osteophyte, capsule changes and joint space narrow in the middle phalanx were 77, 37, 137, 26, 19 and 126 in Ⅱ - Ⅳmiddle phalanx, respectively. Conclusion The Ⅱ - Ⅳ phalanx bones of right hand are the most affected locations in adult KBD.

ABSTRACT

Objective To evaluate the mid-term and long-term effects of early endovascular emboli-zation in treating ruptured posterior communicating artery aneurysm. Methods Early endovascular embolization therapy was carried out in 36 patients with ruptured posterior communicating artery aneurysm (36 aneurysms in total), and the effects of embolization therapy were evaluated by using Raymond grading method. The unified imaging review program was formulated, and the mid-term and long-term effects were evaluated at half, 1, 2, 3 and 5 years after the treatment. Results The technical success rate was 100% in 36 patients (36 aneurysms in total). Raymond grading evaluation showed that gradeⅠwas seen in 32 patients, gradeⅡ in 2 patients, and grade Ⅲ in 2 patients. Recurrence of aneurysm was found in 5 patients (13.89%) at half (n=1), 2 (n=1), 3 (n=1) and 5 years (n=2) after the treatment. Re-rupture of the recurrent aneurysm was seen in one patient (2.78%). No death occurred during the following-up period. Conclusion Early endovascular embolization for ruptured posterior communicating artery aneurysm can effectively improve the prognosis of subarachnoid hemorrhage, and regular imaging examination can promptly detect the recurrence of aneurysm. Effective interventions are helpful for preventing deterioration and rupture of aneurysm.

BACKGROUND:The development of keloid is a progress of fibrosis in wound healing, and involves various fibrosis-related cytokines. Bone morphogenetic protein 7 (BMP7), Gremlin and high mobility group box-1 (HMGB1) play an important role in fibrosis of many organs, but their role in keloid tissue has rarely been reported. OBJECTIVE:To investigate the role of BMP7, Gremlin, vascular endothelial growth factor (VEGF) and HMGB1 in the development of keloid. METHODS:The protein levels and distribution of BMP7, Gremlin, VEGF and HMGB1 in 20 cases of keloid and 20 cases of normal skin were detected by immunohistochemistry and western blot analysis, respectively. And the correlations among expression levels of BMP7, Gremlin, VEGF and HMGB1 in keloid were analyzed. RESULTS AND CONCLUSION:In keloid tissue, the expression levels of Gremlin, VEGF and HMGB1 were significantly higher than that in normal skin (P<0.01), while the expression levels of BMP7 were significantly lower (P<0.01). The levels of Gremlin were negatively correlated with the levels of BMP7 (r=-0.539, P<0.05). And the levels of VEGF were positively correlated with the levels of HMGB1 (r=0.56, P<0.05). The overexpression of Gremlin and decreased expression of BMP7, as wel as the increased expression of HMGB1 and VEGF, may contribute to the pathogenesis of fibrosis in the development of keloid.

BACKGROUND:YY1 is mainly expressed in the undifferentiated epidermic cells in mouse basal lamina, and the expression level is gradual y down-regulated as the differentiation towards suprabasal lamina. The differential expression indicates that, YY1 is one of the regulators in the process of epidermic cells differentiation. OBJECTIVE:To observe the effects of YY1 over-expression on the differentiation of HaCaT cells infected with lentivirus. METHODS:Lentivirus-YY1 was transferred into the HaCaT cells by using Lipofectamine 2000. After selection of the puromycin, monoclonal celllines were established, and the control group were lentivirus-infected HaCaT cells and uninfected HaCaT cells. The expression of YY1 was detected by using western blot analysis. cells in Lentivirus-YY1-HaCaT group and HaCaT-YY1 group were further divided into two subgroups according to the calcium concentration in culture medium, cells were either cultured in low-calcium medium (0.12 mmol/L) for 48 hours, or cultured in low-calcium medium (0.12 mmol/L) for 24 hours and in high-calcium medium (0.35 mmol/L) for additional 24 hours. Keratin K1, K10, K14, and involucrin, filaggrin and loricrin after over-expression of YY1 were detected with western blot analysis. RESULTS AND CONCLUSION:The HaCaT cells were successful y infected with lentivirus-YY1, and we obtained over-expression of YY1 protein in monoclonal celllines under high-calcium concentrations, the over-expressed YY1 could decrease the expression of K1, involucrin and loricrin, thereby preventing the process of epidermal keratinocytes and maintaining the cells in an undifferentiated state. Lentivirus can efficiently infect human immortalized epidermal cellHaCaT, and YY1 may the important factor of inhibiting the differentiation of basal epidermal cells and maintaining the undifferentiated proliferation status.

ABSTRACT

To analyze the expression of bile acid transport correlative proteins in the placenta of patients with intrahepatic cholestasis of pregnancy [ICP]. This case-control study was performed in the Department of Gynecology and Obstetrics, West China Second University Hospital, Sichuan University, Chengdu, Sichuan, China from March 2007 to October 2008. Real time reverse transcriptase polymerase chain reaction was applied for the mRNA expression measurement of 8 bile acid transport correlative proteins, organic anion transporting polypeptide [OATP]1A2, OATP1B1, multidrug resistance protein [MRP]1, MRP2, anion exchanger [AE]2, bile salt export pump [BSEP], multidrug resistance 3, and familial intrahepatic cholestasis [FIC]1, in normal human placentas [n=20] and those with ICP [n=20]. All the transcripts except OATP1B1 and BSEP were detected. Both OATP1A2 and AE2 mRNA were higher while FIC1 was lower in ICP patients. The alteration of bile acid transport correlative proteins OATP1A2, AE2, and FIC1 may be involved in the fetal cholestasis of ICP

Although the etiology remains unclear, the factors of life style and habits may have an effect on the disease.OBJECTIVE:To examine the association of cigarette smoking and body mass index with the risk of clinical benign prostatic hyperplasia.DESIGN: A hospital-based case-control study SETTINGS:Department of Epidemiology and Statistics, Public Health School,Tongji Medical College, Huazhong University of Science and Technology; Department of Preventive Medicine, Liaoning Medical College.PARTICIPANTS:Totally 343 males with benign prostatic hyperplasia of 50-82 years old (patient group),who were surgically treated between May 2004 and May 2006,were selected from the Department of Urinary Surgery of the First Affiliated Hospital and Third Affiliated Hospital of Liaoning Medical College (former Jinzhou Railway Central Hospital),Jinzhou Central Hospital and Jinzhou Second People's Hospital.Inclusive criteria:① males above 50 years old;intemational prostatic symptom score(IPSS)>7;②Digital recta examination (DRE) after miction indicated prostate grade Ⅰ(transverse diameter>3 cm); ③prostatic volume≥30 g;④maximum unnary flow rate<15 mL/s;Meanwhile,361 inpatients with diseases irrelative to the risk factors of benign prostatic hyperplasia,prostatic cancer and prostatitis were selected trom the above hospitals as the control group,they were 50-82 years of age with an average of 65 years old.Informed contents were obtained from all the enrolled subjects.METHODS:All the patients and controls were interviewed using an anonymous questionnaire.All participants were asked to report whether they had smoked 20 packs of cigarettes or more in their lifetimes and,if yes,they were smokers,on the contrary they were non-smokers.Based on usual number of cigarettes reported in the questionnaire, men were categorized as 1-10,11-20,21-30 and >30 cigarettes per day.Based on duration of smoking,the participants were funher classified as never smoke,1-19,20-29 and ≥30 years.Body mass index(BMI)was assessed from measured body mass and body height,and che standards of BMI in adults in China was 18.5-23.9 for normal,24.0-27.9 for overweight and ≥28.0 for obesity),then the participants were further classified as BMI<18.4,18.5-23.9,24.0-27.9 and ≥28.0 subgroups respectively.Years of education(<7,7-11,and≥12) and occupation(worker,peasant, Intellectual and others) were also investigated.The results of the investigation were input into computer to establish database,the odds ratios (OR) and the 95% confidence interval(CI)in relation to the various measures were estimated using unconditional multiple logistic regression models, including number of cigarettes smoked per day,BMI,years smoked,years of education,occupation.MAIN OUTCOME MEASURES:Relationships of number of cigarettes smoked per day,years smoked.BMI,years of education and occupation with benign prostatic hyperplasia.RESULTS:Totally 343 cases in the patient group and 361 cases in the control group were involved in the analysis of results.①Patients smoked≥30 cigarettes per day had an elevated risk of benign prostatic hypertrophy as compared with the non-smokers(OR=1.32,95% CI:0.92-2.58,P<0.01).②The risks of benign prostatic hypertrophy in overweight and obesity were significantly increased as compared with non-smokers(OR=1.68,95% CI: 1.32-3.67;OR=2.35,95%CI:1.83-4.16),③Manual worker (peasant) was negatively correlated with the risk of benign prostatic hyperplasia (χ2=6.62,P<0.01).CONCLUSION:Smoking 30 cigarettes per day was positively related to benign prostatic hyperplasia.For male who were both overweight(BMI 24.0-27.9)and obesity(BMI≥28.0),smoking was significantly associated the increased risk of surgically treated benign prostatic hyperplasia.
